5-Bromo-2-deoxyuridine (brdu) is a pyrimidine analog. Synthetic proliferating cellular DNA is incorporated into the new one in place of thymidine. The Biovision Cell Proliferation BrdU Assay Kit detects incorporated brdu using mouse anti-brdu antibodies. An anti-mouse hrp-bound secondary a is used to detect the antibody test for anti-BrdU-bound BrdU, which is followed by the addition of TMB (an hrp substrate).
The degree of color development is proportional to the amount of brdu incorporated into the cells and can be used directly as an indicator of cell proliferation.
